The flow layer is pressurized constantly during a sorting run (~10 psi), while the control layer can alternate between 0 and ~30 psi. The pressure state of the control layer is determined by the MATLAB code, which controls the open/closed state of the valves in the valve terminal (Festo, S5) via the valve controller (WAGO, S5). When a valve in the valve terminal opens it pressurizes a control layer channel, causing the septum dividing the control layer channel from its associated flow layer channel to distend into the flow layer. This distension acts as a microfluidic valve, blocking flow at that point in the flow layer.
